Peripheral Blood As a Source of Stem Cells for Regenerative Medicine: Emphasis Towards Corneal Epithelial Reconstruction-An In Vitro Study
Background: Mesenchymal stem cell-based treatments are now emerging as a therapy for corneal epithelial damage. Although the bone marrow, adipose tissue and umbilical cord blood is a major source of mesenchymal stem cells (MSC), other tissues such as peripheral blood also harbor stem mesenchymal-like cells called the blood-derived mononuclear cells of peripheral (PBMNCs). This blood derived stem cells gained a lot of attention because of the ease of collection and isolation of minimally invasive. In this study, the feasibility of using PBMNCs as an alternative cell source for corneal limbal corneal epithelial cells regenerate stem envisaging evaluated.
Methods: Rabbits PBMNCs isolated using density gradient centrifugation and evaluated for mesenchymal cell properties including stemness. PBMNCs distinguished for corneal epithelial lineage using conditioned media limbal explants of rabbits and evaluated by immuno-cytochemistry and gene expression analysis. Furthermore, differentiated PBMNCs engineered into the cell sheet using in-house developed thermo-responsive polymer.
Results: The blood cells are derived is shown having properties similar to mesenchymal stem cells. The commitment of the corneal epithelium descendant of PBMNCs confirmed by a positive expression of CK3 / 12 markers that indicate suitability as an alternative to limbal stem cells. Resulting differentiated cells effectively in vitro cell sheets were then shown to a transfer sheet on ex vivo cells excised rabbit eyes.
Conclusion: PBMNCs as autologous cell source alternative to limbal stem cells are considered to be an effective therapeutic strategy for corneal surface reconstruction, especially for patients with bilateral limbal stem cell deficiency.

Human mesenchymal stem cells (MSC) has become a hot topic in the development of cell therapy and biotechnology. All types of MSC stable genome and have the capability of self-renewal. The main source of MSCs is the bone marrow, adipose tissue, umbilical cord and placenta tissue. MSCs can be cultivated in many parts differentiated to grow into cells that are more specialized, produce secretory factor and also supports trophic functions in the body.
Exosomes, derived from MSC also has great potential in regenerative medicine and tissue engineering. Exosomes secreted by MSC and has the same therapeutic potential as the parent cell. MSC and they are combined with a biomaterial exosomes can also be more effective in promoting regeneration of tissues and organs. However, for the use of MSC-exosomes as an agent distinct clinical MSC-exosomes have been produced and their effect therapy shown in clinical studies.
